Do you really mean removing grout or are you referring to the mortar or thinset that bonds the tile to the floor? If it's the latter then you need to scrape it off to make the surface as smooth as possible to bond the new tile. If it's really uneven then you need to float the floor with leveling compound. Also, you should laying cement board down instead of plywood.
