It probably will sense air from behind the drywall, but humidity doesn't respect barriers with cracks, it goes right through them to even the humidity everywhere - as moisture in the air is a gas. The reasong you need them is that house air humidity leaves through all the cracks in the outside walls - windows, door, other openings. The inside wall is surrounded by the air around it in the rooms, so the humidity in that space won't be significantly different unless you can guarantee it is as tight as a diving bell or spaceship.
